2) 80 per cent light to camera and 20 per cent light to eyepiece (for the photography of brightly fluorescing material). (3) 100 per cent light to camera (for the photography of faintly fluorescing material). Fading of the fluorescence during initial focusing can be cut to a minimum by the insertion of a weak grey filter into the light path. It is advisable to focus first, with light at full intensity, on a brightly fluorescing part of the specimen which is not required for photography; major adjustments of the focusing eyepiece can be carried out at this stage.

Provided the same type of film is used throughout, the most satisfactory exposure times for different types of specimens can soon be established. A fast film is necessary, because of the fading of fluorescence, and best results are obtained with colour film. Black-and-white prints can be prepared from the colour transparencies when required and they are usually superior to photographs taken with a black-and-white film, both in clarity and contrast. A 35 mm daylight colour film is generally used, and Kodak Ektachrome 200 (formerly High Speed Ektachrome) and GAF500 (formerly Super Anscochrome) are suitable, if they are specially developed at double the normal rating.

A selection of filter sets for blue-light excitation is offered by the various microscope manufacturers. g. TRITC-1 transmits between 430 and 560 nm); a wide band between 500 and 560 nm or 515 and 560 nm; or a narrow band between 530 and 560 nm with 100 per cent transmission at 546 nm, utilizing the peak transmission of the mercury vapour burner at 546 nm. All these excitation filters are interference filters with a sharp cut-off at 560 nm. The chromatic beam splitter used in conjunction with them reflects light below and transmits light above 580 nm, and the barrier filters recommended are the long pass filters LP580, LP590 or RG610 suppressing light below and transmitting light above 580-610 nm.
