Fading of the fluorescence during initial focusing can be cut to a minimum by the insertion of a weak grey filter into the light path. It is advisable to focus first, with light at full intensity, on a brightly fluorescing part of the specimen which is not required for photography; major adjustments of the focusing eyepiece can be carried out at this stage.
Provided the same type of film is used throughout, the most satisfactory exposure times for different types of specimens can soon be established. A fast film is necessary, because of the fading of fluorescence, and best results are obtained with colour film. Black-and-white prints can be prepared from the colour transparencies when required and they are usually superior to photographs taken with a black-and-white film, both in clarity and contrast. A 35 mm daylight colour film is generally used, and Kodak Ektachrome 200 (formerly High Speed Ektachrome) and GAF500 (formerly Super Anscochrome) are suitable, if they are specially developed at double the normal rating.
