(ii) Use of extrinsic chromophores The approach to measuring rotation of molecules with intrinsic chromophores can, in principle, be generalized by using extrinsic labels. In practice, considerable effort was required to develop a label which was both sufficiently long-lived to allow measurement of protein rotation in lipids and generally reactive with proteins.
Fluorescence polarization methods are quite successful in measuring rotation of labeled proteins in aqueous solutions where the rotational relaxation times (approx. tDrot) are of the order of 10 -8 s, a time about equal to the lifetime of the excited state of the label, 1-100 X 10 -9 s. If solvent viscosity is increased around 100-fold, then near-complete decay of the fluorescence will occur before significant molecular rotation takes place and no depolarization will be detected. In fact, this result (no depolarization) was obtained by Wahl et al.
