Initial experimental studies involving signaling molecules mediated through the pathways have been predominantly performed in a nonconstitutive manner. That is, the two pathways have been studied independently of each other. However, later studies indicated that the two pathways interacted downstream of TRIF and, therefore, may be dependent on each other in the activation of proinflammatory transcription factors NF-κB and AP-1.
